In 12-TET, you make a major third by adding four semitones, which means that you multiply your frequency by the 12th root of 2 four times. That comes to 1.25992, which is noticeably higher than 5/4. Thirds in 12-TET are quite sharp compared to what your ears are expecting from natural harmonics.
